A Study of the Staining Effect of Roselle (Hibiscus sabdariffa) on the Histologic Section of the Testis International Journal of Morphology

This study describes the preparation and use of Roselle (Hibiscus sábddariffa) for the differential staining of testicular tissue sections to find out its staining effect on nuclear, cytoplasmic and other structures. Various treatments using modifications of the plant extract in water were carried out on sections of the rabbit testis. Various levéis of differentiation of nuclear and cytoplasmic structures as well as other structures of this organ was obtained especially when 1% eosin was applied as a counter stain. Contemporary Staining of Arterial System with Cadaver Fixation: An Experimental Study International Journal of Morphology

Although many methods are investigated, cadavers have unique role for education of gross anatomy. This study was performed to make cadaveric based gross anatomy more enjoyable and decrease the time needed for achievement of arterial system for teachers and learners. The adult rabbits in control (n=3) and experimental groups (n=9) sacrificed following deep anesthesia. The samples were fixed by injection of fixative solution to the left ventricle and the corps dropped to the traditional fixative liquid for at least 1 month. Abstract: Seed companies are looking for promising, quick and effective alternatives to determine the physiological quality of seeds. The objective of the current work was to study the efficiency of the exudate - phenolphthalein pH test to evaluate the seeds of two lots of Libidibia ferrea (Mart. ex Tul.) L. P. Queiroz var. ferrea. The statistical design for the the exudate - phenolphthalein pH test was completely randomized with four replicates of 50 seeds in a factorial design (2 x 5), two seed lots and five soaking periods (30, 60, 90, 120, and 150 minutes), respectively, using two constant temperatures (25 and 30°C). On the need for independently estimating organic tripton carbon content OMA

A method is described by which the contribution of Non-Living Particulate Organic Matter (NLPOM) to Total Organic Matter can be determined. The dye Trypan Blue stains only dead organic matter. The surface of the stained particles is used as an indirect measure of the NLPOM-carbon content. Applying this method, it is shown that the bivariate linear regression of POM on chlorophyll <i>a</i> may produce serious errors in the calculated NLPOM contribution when carbon which is neither tripton- nor chlorophyll-related is present in appreciable quantities.

Understanding otolith biomineralization processes: new insights into microscale spatial distribution of organic and mineral fractions from Raman microspectrometry ArchiMer

It is generally accepted that the formation of otolith microstructures (L- and D-zones) and in particular the organic and mineral fractions vary on a daily basis. Raman microspectrometry provides a nondestructive technique that can be used to provide structural information on organic and mineral compounds. We applied it to thin otolith sections of hake in order to address the following issues: (1) the simultaneous characterization of variations in the organic and mineral fractions both in the core area and along successive otolith microstructures; (2) elucidation of significant differences between these fractions; (3) quantification of the effects of etching and staining protocols on otolith structures. Variation in zinc (Zn) concentration in the grain of 50 local rice varieties from Southern Thailand was examined by diphenyl thiocarbazone (DTZ) staining (intense, medium, low, and nil) and chemical analysis. Intense staining was found in the aleurone layer of 94% of the varieties, in the embryo 56% of the varieties and endosperm 12% of the varieties. Variation was found in the DTZ staining intensity of the embryo and endosperm of individual grains, with Shannon-weaver diversity index (H) of the DTZ staining at 0.15-1.00 for the embryo and 0.00-1.00 for the endosperm.
